Yes. As long as the scene has already been tracked and exported from After Effects, you can build, adjust, and prepare the scene directly in Blender. This makes collaboration easy between artists using different tools.

If you’re doing the tracking yourself, then yes — you’ll need After Effects to run the built-in 3D Camera Tracker and export the scene data. However, if someone else is handling the tracking part, you don’t need After Effects at all. You can simply use the exported .json file and work entirely inside Blender.
